Before the race started, it rained heavily again in Assen following a wet FP3 and Superpole.
But that didn't last long, as blue skies arrived just as the race was about to start, further confusing the riders and teams.
However, while most riders opted for slick tires, Nicolas Spinelli made a big impact early on and decided to go for intermediate tires.
Danilo Petrucci's replacement took the lead early on and cleared it when Sam Lowes jumped into the pit lane at the end of the first lap.
By the end of the second lap, Spinelli had built a 13-second lead as Andrea Locatelli, who was also running at the intermediate front, began to move forward.
Spinelli continued to extend his lead by over 24 seconds over Axel Bassani, but the track was rapidly drying out.
Andrea Iannone made a mistake when trying to keep up with Alvaro Bautista's pace and high-sided his Panigale V4 R machine at the start of Turn 2.
After seven laps, Spinelli's advantage on intermediate tires disappeared as Bassani, Razgatlioglu and Bautista lapped faster.
Bautista then followed Razgatlioglu and overtook Bassani, setting the fastest lap of the race.
A mistake by Razgatlioglu at the beginning of lap nine allowed Bautista to move into second place at Turn 1.
With nine laps to go, Spinelli's lead was down to 11 seconds, and Bautista and Razgatlioglu began to drop off the trailing group.
At the end of lap 14, Razgatlioglu made a nice move on Bautista to take second place, but Spinelli quickly moved into first place a few corners before the corner.
However, more drama ensued when a red flag came out just over two-thirds of the way through the race distance, and Spinelli took victory in his first Superbike race.
A red flag was waved due to oil released onto the circuit by Locatelli's Yamaha.
