Coventry striker Ellis Sims scored a hat-trick in the first half to end Maidstone's dreams of reaching the historic FA Cup quarter-finals.
The sixth-tier Stones, who have already sent out Barrow, Stevenage and Ipswich from the English Football League, were aiming to become only the second non-league team in 109 years to reach the last eight.
They played against the Sky Blues, who were still feeling the effects of Friday's Championship defeat at home to Preston, but were sent off by the 35th minute and Casey Palmer, back on his return, contributed to all three of Sims' goals.
Fabio Tavares added two more goals late in the game and Coventry reached the quarter-finals for the first time since 2009.
After scoring the opening goal, Sims held up a shirt with teammate Tatsuhiro Sakamoto's name on it, but he was injured in that game against Preston, with manager Mark Robins taking him to hospital with a “really bad injury”.
